Slavia could face teams like PSV Eindhoven, Leicester, Fenerbahce, Olympique Marseille, Midtjylland, Celtic or Rapid Vienna.
It is certain that Leten will play the first match at home, while Susians will play the return leg in Eden.
The European Conference League knockout matches are scheduled for 17 and 24 February.
